You actually enter one of the rooms on transition or you get stuck if you're not aligned to slide into one. If you enter the bottom right tile you'll stand on the spikes or blocks and can walk into the room (and fight Mothula). The spikes won't hurt you when standing on them but they will if they're about to move.
